High Throughput Peptide Synthesis Core Facility

Overview of Services

The UNC Peptide Synthesis Core Facility offers a full spectrum of services for custom peptide production, chemical modification, purification, and analysis of synthetic peptides. Our services are open to both UNC investigators and external researchers. Our core facility routinely synthesizes peptides up to 40 amino acids, utilizing automated, microwave assisted Fmoc Solid Phase Peptide Synthesis (SPPS), but can make longer sequences when needed (up to ~80 residues). Flexible synthesis scales (10–300 µmol) allow for quantities ranging from a few milligrams to hundreds of milligrams of crude peptide. Researchers can choose from a wide array of chemical modifications, including post-translational modifications such as acetylation, methylation, propionylation, crotonylation, butyrylation, and phosphorylation; fluorescent dyes and biotin labeling; PEG spacers; incorporation of D-amino acids, unnatural amino acids, and stable isotopes labeled amino acids; as well as various cyclization strategies and metal-chelating tags. Specialized products include C-terminal peptide hydrazides for Native Chemical Ligation and Multiple Antigenic Peptides (MAPs) for antibody generation. The facility also offers synthesis of mid-size peptide libraries (approximately 100 peptides per library) and walk-up MALDI MS instrument access for trained UNC users. With extensive experience in histone peptide synthesis - including PTM-modified, fluorescent, and biotinylated variants -the core supports applications in antibody production, enzyme assays, ligand-binding studies, and quantitative biophysical analyses such as NMR, ITC, and crystallography. By combining technical expertise with flexible service options, the UNC Peptide Synthesis Core ensures high-quality peptides tailored to diverse research needs.
