The Quantitative Targeted Proteomics (QTAP) Core specializes in the quantification of proteins in biological matrices by LC-MS/MS. The core is newly established (2024) but the laboratory has been working on targeted proteomics for over fifteen years.
The key sequential steps in the operations of the core are
1) Extraction of proteins from samples
2) Enzymatic (e.g. trypsin) digestion of the proteins
3) Analysis of the digests by LC-MS/MS
4) Analysis of the data generated
The third step is the most important with respect to core activities as the other steps can be completed outside the core.
Quantifications can be undertaken employing labeled standards (more accurate) or it can be label free (less accurate or relative). The analytical techniques employed by the core are nano- or microLC coupled to triple quadrupole mass spectrometry.
